Trying Spermidine for Autophagy – Thoughts on Dosage and Timing?

I’ve been following the hype around spermidine because the idea that a naturally occurring polyamine could kick‑start cellular clean‑up sounds too good to ignore. I’ve read that people usually take 1‑10 mg a day, usually once daily, and that it can be in supplement form or through food like wheat germ, aged cheese or natto. I’m not a chemist but I reckon the 3‑5 mg range would be a safe start for me, given the short half‑life and low side‑effect profile. I’m a bit cautious because the long‑term human data is still developing, and I’ve had mild gut upset before with other supplements.
I want to ask: would you recommend taking it with a meal to minimise GI upset, or is an empty stomach better for absorption? Also, is there a benefit to cycling (e.g. 3 days on, 4 days off) or should it be daily? I’m thinking of pairing it later with NMN, but I want to keep the protocol simple for now. Any personal experience or insights would be highly appreciated.

- kayla_s: For me, taking spermidine with food made the GI stuff less of a bother, but I’m not a scientist so who knows. I started at 3mg daily with breakfast, which felt fine, no immediate gut issues. Cycling? I’ve seen people do it to “reset,” but I just kept it daily because the hassle of cycling didn’t feel worth it for something that seemed low-risk. If you pair it with NMN, I’d guess timing might matter, but I’ve never done that combo.
- greg208: Thanks for the heads‑up, kayla. I’ll start the 3 mg with breakfast just like you and stick to daily dosing for now – the cycling hassle does feel overkill if the risk is low. If you ever pair NMN, what time did you take it relative to the spermidine? I’m curious whether the same meal helps keep the gut settled.
