Rapamycin and metformin together, is anyone actually running this combo?

Right, so ive been digging into the longevity side of things for a while now and keep coming back to the same two names. Rapamycin for the mTOR/autophagy angle and metformin for the AMPK stuff. Both have proper human data behind them, both are on the profile pages here, and the logic of stacking them seems solid on paper since theyre hitting different pathways.

Heres my question though. Is anyone here actually running both at the same time, and if so whats the schedule looking like? My head says rapamycin pulsed once a week makes sense because of the long half life, but metformin daily. Im not sure if thats overkill or if the pathways actually complement each other well enough to justify it.

Im based in the UK so getting either prescribed off label is a bit of a faff, GP isnt going to play ball for this sort of thing. Not gonna lie, the telemedicine route keeps getting mentioned but i havent pulled the trigger yet.

If youve got experience with either on its own id love to hear how you got on, especially around sides. The mouth sore thing with rapamycin sounds grim and the GI stuff with metformin is enough to put me off honestly.

  • recovery_freya: Cant speak to the combo myself, im only running rapamycin on its own right now, pulsed once a week at a low dose. Metformin is on my list to add eventually but I wanted to see how one variable moved my numbers first before stacking. That’s just how I like to do it. On rapa solo, the mouth sore thing is real but it hit me more like mild canker sores than anything dramatic, cleared up after week two.   • greg208: Cheers for the reply, that actually makes sense what you said about moving one variable at a time, bit daft of me not to think of it that way. I might do exactly that and start with rapa solo first, get a baseline set of bloods, then layer metformin in once ive got something to compare against. Did you do fasting bloods or did your GP run a full metabolic panel with lipids and inflammatory markers thrown in?
  • fasted_scientist: Fair enough on the single variable thing, but when does that logic end? People stack 10 things and always say "I'll add one at a time" but somehow they're always adding. Not having a go, just reckon most of us end up doing the same thing eventually. On the lipids though, what sort of nudge are we talking? Mine went a bit sideways on rapa so now I'm watching it closely. What was your dose?
  • greg208: Not gonna lie you caught me out there. Im definitely guilty of that slow creep of adding stuff one by one. I havent actually started the rapa yet so i dont have any recent bloodwork to show you. Im planning on starting with a small 5mg dose once a week to see how i feel. Sorry i didnt clarify i was still in the planning stage. Have you noticed any specific way your lipids shifted when yours went sideways?
