Saw this MS News Today piece about dasatinib and quercetin, the classic senolytic combo, causing pretty severe myelin loss in lab studies. The researchers basically showed that the drug cocktail destroyed oligodendrocytes, which are the cells that make myelin in the first place.

Honestly this is the kind of finding that makes me glad I never jumped on the D+Q bandwagon. The whole senolytic space feels like it's moving way faster than the safety data, and most people I see taking it are doing it off very little human evidence. Lab models aren't people, sure, but losing your myelin sheath is not a small side effect to wave off.

Anyone here been tracking senolytic research closely? Curious if anyone knows if oral dosing routes show the same CNS penetration, or if this is more relevant to people exploring higher-dose or localized protocols. πŸ₯—

  • grinder265: I read that piece too, and honestly it gave me pause. The oligodendrocyte thing is rough because those cells don't regenerate quickly at all. For me I've always been leery of senolytics because the long term human data just isnt there yet. D+Q sounds clean on paper but youre still messing with apoptosis pathways systemically, and ngl that has downstream effects nobody can really map yet. If it's only at supraphysiological concentrations it's basically a toxicology note and not a practical warning, but if it scales with dose then anyone running higher oral protocols should probably pump the brakes for now. I haven't seen the full paper yet, just the summary, so I'm not sure what doses they used. If you find it let me know, I wanna pull the actual numbers. I didn't see them break it down clearly in the writeup, but I'd guess the concentrations they used were on the high end since they were trying to show a mechanistic effect. Still, even at supraphysiological doses, if oligodendrocytes are getting wiped out, that tells you something about the target itself. Not super comforting πŸ˜…
