Trying low-dose rapamycin weekly for 6 weeks, what should I watch?

Okay so I finally pulled the trigger and started low-dose rapamycin about a month ago, once a week. I'm mostly interested in the autophagy angle and the immune rejuv stuff that keeps coming up in the longevity space.

Background on me: late 30s, generally healthy, bloodwork decent, training 3-4x a week. Not on anything else right now apart from creatine and fish oil. No meds, no other peptides.

So far the biggest thing I've noticed is honestly just feeling a bit "quieter" in my body on the dose day itself, kind of a low energy thing that fades by the next morning. Mouth was a tiny bit sore around week 3 but nothing major. Sleep has actually been pretty solid.

What I want to ask the room: what are you guys actually tracking when you run it? Like beyond the obvious lipid panel, is anyone getting fasting insulin done, hsCRP, or any of the newer immune age markers? And how long before you saw anything you'd call a real change?

Also curious if anyone pairs it with anything or just runs solo. Not looking for a protocol, just want to compare notes before I check in with my doc for repeat bloods 😅⚡

  • weekendnate: I tracked fasting insulin and hsCRP at baseline and again around week 6, plus a standard lipid panel and a CBC with differential. Honestly the fasting insulin was the one I cared most about because mTOR signaling ties into insulin sensitivity. For me the lipid changes showed up faster than anything else, but that could just be noise from the timing of the dose.   • elle_z: Honestly the fasting insulin angle is what I was leaning toward too, my baseline was already kinda meh so I want to see if it actually moves. Did yours change much at week 6 or was it more of a slow drift? I haven't done a CBC with diff yet, gonna add that to my next draw. And yeah the quiet feeling did ease off a bit by week 4, figured it was just my body adjusting but it's nice to hear yours did the same.
  • elle_z: The mucositis thing is good to know is common, I was wondering if I was imagining it. Did you do anything for it or just wait it out? And yeah the fasting insulin angle makes sense, I should probably get that added to my next blood draw lah instead of just waiting.
