Found this piece on peptide cycling and protocol design. It goes into how researchers think about receptor dynamics, on and off periods, and why blindly running something for 12 weeks then stopping cold is a bit naff.

The bit on tachyphylaxis and receptor downregulation was decent. That's the bit most bro-science stacks completely ignore. People just run the same thing forever then wonder why it stops working.

The one thing I'd push back on is it leans a bit textbook. Real world a lot of us aren't designing trials, we're messing about with BPC for a niggle and seeing what happens. Curious if anyone here actually structures their cycling off any of this logic, or if it's more vibes and bloods...

  • quiet_zone2: I’ve played around with cycling a few peptides myself, mostly BPC‑157 and a bit of CJC‑1295 for recovery. For me the sweet spot seemed to be about six weeks on then four weeks off – I noticed the soreness relief faded after week five and came back stronger after the break. I didn’t run any formal receptor tests, just paid attention to how my joints felt and did a basic blood panel before and after each cycle to make sure nothing was drifting. It’s more vibes and bloods than a strict protocol, b
  • nightryan: Tbh the 6 on 4 off pattern matches what most people i know land on too, week 5 or 6 is usually where things go flat. I do something similar but i track HRV and sleep score daily, way more useful than just pain scores imo because you catch the drop before you actually feel it. Bloods are smart though, what were you pulling each time?
  • humbleomar229: Honestly yeah the week 5/6 flatness thing is what got me thinking about it in the first place. I wasn't pulling anything fancy, just standard bloods before and after a cycle, lipids, liver, the usual. Nothing dramatic came back flagged either time but i wasn't tracking HRV at all so i had no clue if my body was actually coping or just quiet about it... That's a decent shout, might set something up next round and actually compare the two.
  • humbleomar229: Yeah six on four off is roughly where i landed too, though mine was more like five on six off because i'm paranoid about the receptor thing. Vibes and bloods is basically what we have tbh. No one here is running proper assays, we're all just guessing based on how our dodgy knees feel on a tuesday morning... Symptom logging is a good shout though. I've been half arsing it with notes in my phone but nothing structured. Reckon an actual log would help separate real signal from placebo. What did y
  • humbleomar229: Cheers for that, the week five fade is exactly what I'd expect if downregulation is doing its thing. Six on four off sounds reasonable... I haven't tried formal symptom logging yet tbh, just rough notes in my phone when something flares up. Did you notice the CJC side giving you any water retention or weirdness, or was it pretty clean?
