12 weeks on tirzepatide, considering adding amlexanox for the last stubborn 8 lbs

Been on tirzepatide since April, currently at 12.5 mg weekly. Down 42 lbs and my bloodwork looks the cleanest it has in years. A1C went from 5.9 to 5.2, fasting insulin dropped, lipids all moved in the right direction. Tracking calories, lifting four days a week, walking 8 to 10k steps daily. The last 8 to 10 lbs though are playing hardball. Hunger is mostly gone, training is dialed in, but the scale barely moves week to week anymore.
Started reading more about amlexanox and the TBK1/IKKe angle. The mechanism makes sense on paper, basically trying to flip the metabolic thermostat back up when GLP-1 signaling plateaus. A friend in a similar situation tried it for six weeks and broke through a multi-month stall without crazy side effects, just some mild GI stuff early on.
For those who have stacked it on a GLP-1, how did you actually time it around your shot day? And did anyone notice any weird interactions with HRV or sleep? I track both with a Whoop and like to keep an eye on trends before adding anything new. Curious what the real world experience looks like before I bring it up with my doc.

- busy_dad: Congrats on the progress, that bloodwork swing is impressive honestly 📚 I have not stacked amlexanox with a GLP-1 myself but I have read a bit about the TBK1/IKKe angle and the rationale does line up with what people report when they hit a wall on these drugs. A couple things I would watch hard before pulling the trigger. HRV and sleep are smart metrics to anchor on, since any inflammatory shift can show up there before you feel it. Did your friend's Whoop numbers move at all during those six
- weekendnate: Will check with him, but I do not think he is a Whoop guy, more of a scale and weekly photo type. I run an Oura ring and my HRV has been flat for the last three weeks, which is one reason I am considering this. Morning versus evening is a, I have seen people mention splitting it to avoid the GI dip. Did you time yours a certain way, or were you just guessing?
- runner_aaron: Hey, solid reply from busy_dad there. Tbh the HRV angle is a good shout, I wouldn't have thought to anchor on that myself. Did your friend log any morning vs evening difference? Also was it oral amlexanox the whole time or did they mess with timing around food?
- weekendnate: I think they stayed oral the whole time, 50 mg split AM and lunch with food since the bioavailability story looked worse fasted. No HRV data I saw, just weight and a food log, so I can't really speak to morning versus evening beyond what I noticed on myself. I started yesterday, dosing with meals, and my resting HR is up about 4 bpm versus last week which I'll keep an eye on. Will log it for two weeks before deciding if it actually does anything.
