MK-677 fasting glucose creep, how long before it actually matters?

Hey. I have been on 25mg MK-677 nightly for about 6 weeks now, mostly for the sleep and recovery side of things, and my fasting glucose has been slowly climbing. Started around 5.2, last test showed 5.9. HbA1c still in range but trending the wrong way. I know the glucose thing is a known side effect, I just did not expect to actually see it on paper this quickly.

Few things I am wondering about tbh. Does it settle if you push through, or does it keep creeping the whole time you stay on it? And has anyone tried splitting the dose or dropping to 12.5mg and still kept the sleep benefits? The deep sleep was the main reason I started, so I do not want to lose that part.

Also, does timing of the dose matter for the glucose angle, like taking it earlier in the day with food vs on an empty stomach at night? I have been doing it right before bed, no food. Not sure if that is making it worse or if it makes no real difference.

Bloodwork is the one thing keeping me honest here. Would rather catch it early than pretend it is not happening.

  • grinder265: Yeah the glucose creep on mk-677 is real and from what ive seen it doesnt really settle, it kinda tracks with how long youre on it. 6 weeks is still pretty early too so ngl i wouldnt expect that to be the peak. Splitting the dose didnt do much for me when i tried, and 12.5 was too weak for sleep. The deep sleep benefit felt pretty dose dependent for me. Timing though, i moved mine to right before dinner instead of bedtime and the morning fasting numbers came down a little. Not huge but noticea
  • runner_aaron: That is what I was worried about tbh. If it just keeps climbing, I cannot stay on 25mg for long. 9 is already pushing it for me. I will try moving the dose to before dinner like you did and see if those morning numbers drop. Do you feel like the sleep quality took a hit when you stopped taking it right before bed?
  • tiredmira: The timing swap is interesting actually. 2 by week 8 or so. I never thought to move it earlier in the evening. That tracks though, less overlap with the early morning cortisol/insulin window probably. Did you notice the sleep quality dip at all when you moved it, or was it basically the same? For me the deep sleep effect was kinda fragile so I am curious if that held.
  • runner_aaron: Yeah the sleep was basically the same tbh. Maybe took an extra day to fall asleep the first 2 or 3 nights but the deep sleep stuff came back quick. Fasting glucose dropped from 5.9 back to about 5.4 over the next couple weeks so something shifted.
