Semaglutide slowed ageing in old female mice, the longevity crowd is losing it

Nature just put out a study showing late-life semaglutide treatment extended lifespan and slowed ageing markers in female mice. Not males, just females for now, and the effect size on lifespan was pretty meaningful for a single drug started that late in life.
Honestly this is the most interesting longevity paper I've seen in a while, even if it's still mice. Most of what gets hyped in the anti-age space is either senolytics with shaky human data or supplements that probably do nothing. A GLP-1 agonist pulling real lifespan extension in aged animals is a different kind of signal.
The obvious question is whether the lifespan bump is from weight loss and metabolic improvement, or something more fundamental going on in the cells. I'd love to see them tease that apart. Also can't wait for the male cohort data, because right now half the population is being left out of the interesting finding.
Anyone else following this one closely or think the media is overreading it?

- fasted_scientist: Ngl this one caught my eye too but I'm trying not to get carried away. Mice living longer on a GLP-1 isn't really new information, we already knew caloric restriction and metformin-adjacent stuff shifts lifespan in rodents, and semaglutide makes them eat less so are we actually surprised? The interesting bit for me is the late start. Most longevity mouse work begins treatment young, so seeing benefit when you intervene in already old animals is the actual signal worth chasing. I'd want to see
- elle_z: Yeah exactly that "started treatment late" point is what made me stop scrolling too. Most of the mouse longevity stuff starts at like 12 months equivalent and yeah of course things live longer, give them less food and they live longer, we knew that. Starting at old age is the part that actually matters if anyone is hoping this translates to people who are already past middle age. And lol fair on me losing the thread at the end, my phone did something weird, was saying the male cohort data can't
- runner_aaron: Yeah that late start thing is exactly what jumped out at me as well tbh. Most mouse longevity work kicks in early, so starting at an age where they're already showing decline and still pulling meaningful lifespan is the part I'd want to dig into more. Fair point on the eating less angle though, hard to untangle a direct drug effect from just reduced calories when the mice are voluntarily eating less anyway. Curious if they controlled for that at all.
- elle_z: Honestly not 100% sure from the abstract whether they pair-fed the control group, that's the one methodological thing I want to check when I get a chance to read the full paper properly. Pair-feeding would at least separate drug effect from just "ate less, lived longer" which we kinda already know happens in mice 😅
