Metformin before bed vs with dinner, does timing actually matter for the longevity crowd?

I've been on low-dose metformin for about eight months now, mostly for the AMPK and longevity angle since my bloodwork has been solid and I wanted to get ahead of the metabolic stuff we see creeping up in our 30s. Something I've been going back and forth on is timing, I've tried both with my biggest meal and right before bed, and I honestly can't tell if it's making a real difference or if it's just whatever fits my schedule that day.
When I take it with dinner I sometimes get that mild GI thing if I've pushed my carbs higher, but moving it to right before bed seems to make mornings feel a little cleaner energy wise, could just be placebo though. I've also wondered if there's any logic to hitting AMPK overnight when you're already in a fasted state versus tagging it during a meal when your body is processing glucose.
For anyone here who's been on it longer, did you settle into a timing that felt best, and did you notice anything with fasting numbers or training recovery shifting one way or the other? Genuinely curious if the longevity folks have a consensus or if it's just whatever your stomach tolerates ðŸ§

- grinder265: Ngl the longevity crowd doesn't have a real consensus on this, mostly just personal preference and some gastro tolerance reasoning. From a pharmacology angle, the half-life of metformin is only about 6 hours so timing doesn't really matter for sustained AMPK activation the way people think it does. You're not hitting some overnight window, it's just circulating either way. The before bed thing came from older T2D studies showing marginal improvements in fasting glucose, not from longevity resea
- patient_codes: Yeah the half-life thing makes sense, I had been assuming it was hanging around longer than it actually is lol. The "overnight AMPK window" framing was kinda my own mental model and you're right it's probably more about when it's easiest on my stomach than anything else. I'll go back to taking it with dinner and stop overthinking the timing 🧠thanks for breaking down where that idea actually came from, that fasting glucose note from the T2D studies tracks.
- tiredmira: Yeah the half-life point kinda kills the whole "overnight AMPK window" argument for me too 😅 I run mine with dinner and my fasting numbers are basically identical to when I did the before bed stint. Basically just comes down to which one your stomach tolerates better.
- patient_codes: Yeah that lines up with what I saw honestly, the half-life thing is a good call out because I kept telling myself the overnight angle was doing extra work but it probably isn't doing much past a few hours. GI stuff is the only real differentiator for me too, going back to dinner since mornings were a wash anyway, thanks for the reality check ðŸ§
