Low-dose rapamycin for 8 months, what i actually noticed

To be fair i was on the fence for a long time before pulling the trigger on rapamycin, the immunosuppressant angle scared me more than anything. But after reading through the longevity research and talking to my doctor i started at 3mg once a week, bumped to 5mg after the second month. Bloodwork every 8 weeks, lipids, fasting glucose, the usual stuff.

What i actually felt: around week 6 i noticed my recovery after heavy training shifted, less sore, less wiped the next day. Sleep got a bit deeper too, though that could just be placebo. Skin looked fresher to my eyes but my partner politely disagreed so take that with a grain of salt 👀

Side effects: one mouth sore around week 3, gone in 4 days. Lipids went up mildly at the 3 month mark then settled back down. Nothing major, but i keep an eye on it.

Biggest thing for me is the autophagy idea, that cellular clean-up stuff. Hard to measure from the outside but i like the theory and so far my bloodwork hasnt given me a reason to stop.

Anyone else been on it longer than me? Curious how the 12 to 24 month markers looked for you. âš¡

The recovery thing is what caught my eye, mine has been fine lately but im still early so im watching that one closely. Mouth sores sound rough, glad it cleared up quick for you. My bloodwork hasnt shown anything weird yet either but im only a few weeks in so take that with a pinch of salt. Did your doc say anything about the lipid bump, like was there a plan if it kept climbing, or did you just ride it out?
  • mateo_w: To be fair my doc wasnt super worried since it was mild and settled on its own by month 4. He basically said keep monitoring and if fasting glucose or ldl kept trending up at the next draw wed reassess. Since i have a training background he also mentioned keeping protein and fibre high, which i already do mostly. Riding it out worked for me but yeah having a plan b if it climbed further was reassuring.
  • katie_sauna183: The lipid bump was the part that made me nervous too when I started. My doc said we would recheck in 8 weeks and if triglycerides kept trending up we would discuss dropping the dose or adding a statin, but for me the numbers settled around month 4 without changing anything. Definitely keep an eye on it though, lipids creep up slowly and you don't feel it happening.
  • mateo_w: Yeah that matches what mine did, it creeped up at the 3 month bloodwork and i was ready to bail honestly. By month 5 it was actually back under my baseline from before i started, weird. Maybe the body adapts since my diet and training didnt change. Doc said same thing yours did, recheck then decide, so we are on the same page there. Triglycerides or LDL specifically, or just total cholesterol you saw move?
