PE 22-28 and TREK-1 blockade, is anyone else watching this space?

I have been going down the rabbit hole on PE 22-28 lately. It is a spadin-derived peptide that blocks the TREK-1 potassium channel, and the preclinical angle is fast-acting antidepressant effects plus neurogenesis. You can read the profile here: /peptides/pe-22-28.
Honestly what caught my attention is the mechanism. TREK-1 is a background potassium channel, so blocking it basically makes neurons more excitable and pushes serotonergic signalling. The idea that you could get something that acts faster than SSRIs is appealing on paper, but I keep reminding myself this is all animal data so far. No human safety data, no established dosing. That part makes me cautious even though I am usually the first to try things.
I have not tried PE 22-28 myself. The lack of any human pharmacokinetics data is honestly the thing that stops me. A short half-life peptide with unknown human metabolism is not something I want to guess at.
What I find promising though is the neuroplasticity angle, not just mood. If TREK-1 blockade genuinely increases synaptogenesis, there could be interesting applications beyond depression. Stroke recovery research is mentioned in the profile and that seems worth tracking.
