"My first FOXO4-DRI senolytic cycle – what I felt over three weeks"

I decided to try a short FOXO4‑DRI run after reading about its senolytic mechanism. I used a very low dose, injected subcutaneously twice a week for three weeks, just to see how my body reacted. Honestly the first few days I felt a bit of low‑grade fatigue and a mild headache, nothing that stopped me from work.
By day five the fatigue faded and I noticed my usual morning brain fog seemed a little lighter, though I can’t say if that was the peptide or just better sleep. My skin felt a tad smoother on the forearms, but again that could be placebo. I kept a simple log of energy, mood and sleep scores each night.
After the three weeks I took a break of two weeks before starting any other compound. I’m curious if anyone else has tried a similar low‑frequency FOXO4‑DRI protocol and what markers they looked at, like CRP or WBC, or if they stacked it with something like epithalon for telomere support.

- curious_scientist: I’ve only done a single low‑dose FOXO4‑DRI run myself, twice a week for four weeks, and I noticed a similar dip in energy the first couple days then a subtle lift in afternoon focus that faded after I stopped. I didn’t run any labs but I tracked resting heart rate and felt my recovery after light PT sessions was a tad quicker, though that could just be better sleep hygiene. I’m also curious about stacking – did anyone pair it with something like epithalon or a low dose of rapamycin and see addit
- sleep_coastal: Anecdotally I tried FOXO4‑DRI once with a low dose of epithalon (5 mg subQ twice weekly) and didn’t see any clear change in my resting heart rate or perceived recovery beyond what I got from the peptide alone. No labs, just symptom tracking. Curious if others noticed anything different.
- sleep_coastal: I stacked FOXO4-DRI with low-dose rapamycin on one cycle, alternating days not same day. Anecdotally the afternoon focus lift felt a bit more sustained but I can't say it was additive versus just the rapamycin doing its thing. I did get CRP checked before and after and it dropped slightly, nothing dramatic. WBC stayed in range. Could've been anything though.
- midwest_scientist: I didn't get bloodwork done before mine which honestly was a bit stupid of me. Did you run the rapamycin at like 3 to 5mg weekly or something lower? I'm wondering if the alternating day approach matters much versus just spacing them a few days apart.
- midwest_scientist: That is interesting about your resting heart rate. I didn't track my heart rate during my three weeks, I only looked at energy and mood. It sounds like the combination didn't do much extra for your recovery. Did you notice any changes in skin or sleep quality with that epithalon dose, or was it completely neutral for you?
- midwest_scientist: The afternoon focus fading after you stopped matches what I would expect from a short senolytic pulse, honestly that lift is subtle enough it could just be the fatigue clearing. Your resting heart rate tracking is a good idea, I wish I had done that instead of just subjective scores. Did you track RHR with a watch or just manual? I am also very curious about the rapamycin angle, that is next on my list to look into before my second cycle.
