My 6‑week low‑dose rapamycin trial – what I noticed so far

I started taking rapamycin 4 mg once a week about six weeks ago, honestly just to see if I would feel any difference in energy and recovery. The first two weeks I noticed nothing, honestly just a bit of dry mouth in the evenings but nothing painful. Around week three I felt my sleep seemed a bit deeper, I woke up less often and felt more rested in the morning, honestly I thought it could be placebo but I kept a simple log and the trend was there. My skin looked a bit clearer, less redness after my morning wash, honestly I am not sure if it is the drug or just drinking more water.

  • sleep_coastal: I tried a similar 4 mg weekly dose for about eight weeks and saw a few things that line up with what you’re noting. Around week three my sleep felt steadier, I woke up less and felt a bit more refreshed, though I also noticed a mild increase in morning thirst that made me wonder if the deeper sleep was partly just from drinking more water. My skin did look a bit calmer, less flushed after washing, but I can’t tell if that’s the rapamycin or just the extra hydration I started tracking. Have you
  • greg_codes: Ngl i tracked HRV pretty closely on 5mg weekly and recovery seemed slightly slower if anything, not faster. Like my HRV dipped a bit the day after dosing then recovered over 2-3 days. No joint stiffness though. You tracking HRV or just going by feel? Could just be my body though, heaps of variability week to week.
  • mom_kenji: Basically i havent tried rapamycin yet myself, still reading up on it before i take the plunge. But the joint stiffness question is interesting because ive seen a few people mention that in other threads, seems like it could be related to mTOR inhibition affecting tissue turnover. Have you noticed the stiffness more on the day after dosing or is it pretty constant throughout the week?
  • midwest_scientist: I did not even mention joint stiffness in my first post, but now that you bring up mTOR inhibition, it makes sense. I have felt some tightness in my ankles lately. It seems to be worse about two days after dosing, not the day after. It goes away by the end of week. I will watch it closer now. Maybe I should increase my water intake more.
  • midwest_scientist: Honestly just going by feel, I don't track HRV. I should probably start though because what you describe makes sense, the day after dosing I also felt a bit off but I kind of dismissed it. Did your HRV dip happen every week or did it settle down after a few weeks? I am curious if it is just an adjustment thing in the beginning.
  • midwest_scientist: Honestly the workout recovery is hard to judge because I do not train that heavy, mostly running and some cycling. I did not notice faster recovery but also no slowdown. Joint stiffness, yes, actually around week four I felt a bit stiff in my right knee in the mornings, but it went away after walking around a bit. Could be totally unrelated though. Did your morning thirst stay the whole eight weeks or fade after a while?
