GlyNAC vs glutathione – what are people actually feeling on daily dosing?

I’ve been experimenting with glynac for about six weeks now, splitting the dose morning and evening as suggested (glycine ~100mg/kg + nac ~100mg/kg). For me it seemed to smooth out some of the afternoon fatigue i usually get after long runs, and my sleep felt a touch deeper – though i’m not sure if that’s just the extra rest days i’ve been squeezing in. I’ve also tried a low‑dose glutathione IM shot once a week for comparison, and that gave me a brief brightening in skin tone but didn’t really shift my energy or recovery noticeably.

I’m curious if others have run both and noticed a clear difference in day‑to‑day feeling, or if the effects are more subtle and only show up on bloodwork. Any thoughts on dosing frequency or stacking with something like rapamycin? Keen to hear what’s working for you all. Chur.

  • optimizer_data276: I tried glynac for about two months at a similar dose and honestly the effect was pretty subtle for me too, mostly just less next-day soreness after hard training days, could've been placebo or just better sleep habits around that time. The glutathione IM route is interesting because orally it gets chewed up pretty fast by the gut, so IM would bypass that, but i'd guess the skin brightening you noticed is more of a short-term cosmetic thing than a real systemic antioxidant shift. On the rapamyci
  • student748: Yeah nah that tracks with what i'm seeing, the glynac effects are pretty subtle for me too. The next-day soreness thing is interesting though, i didn't notice that specifically but maybe i wasn't paying enough attention to it. For what it is worth the skin brightening from the IM shot did fade after a couple days so you're probably right about it being cosmetic rather than systemic. On the rapamycin stacking, i'm already doing rapa 5mg weekly so maybe i should just get proper bloodwork before a
  • fasted_scientist: I reckon the GlyNAC thing is interesting because the mechanism actually makes sense on paper, giving the body the precursors to make its own glutathione rather than dumping it in exogenously. But ngl I'm always a bit sceptical when people say they feel precursors working in a few weeks. Could just be the glycine improving sleep quality on its own, there's decent data on glycine and sleep. The IM glutathione giving you a skin effect but not much else sort of tracks with what I've seen people rep
  • sleep_coastal: I actually ran a basic GSH panel after six weeks of split‑dose GlyNAC. Total glutathione rose about 15% and glycine alone did improve my sleep latency, but the energy boost felt more than just glycine. No dramatic changes, just a subtle shift.
  • fasted_scientist: Ngl I reckon you are spot on about the gut issues with oral glutathione. But does anyone actually have a study showing that the skin brightening is just cosmetic? I am keen to see if there is actual data on that or if we are all just guessing based on how it looks in the mirror.
  • student748: Yeah nah that's a fair call, i don't actually have any data on whether the skin brightening from the IM shot is doing something systemic or just cosmetic. For what it is worth the effect i noticed faded after a few days so it could just be transient. Would be keen to see if anyone here has tracked oxidative stress markers alongside it, that might tell us if it's actually shifting something real or just surface level.
  • sleep_coastal: I ran a basic CBC and glutathione levels before and after six weeks of GlyNAC. GSH rose about 15% and my fasting glucose felt steadier. Glycine alone helped my sleep, but the combo seemed to add a bit more daytime clarity.
  • student748: Oh nice, actually getting bloodwork before and after is smart as. That 15% GSH bump is interesting, did you get a sense of whether the fasting glucose change held up beyond the six weeks or was it more of a short-term thing? For what it is worth the daytime clarity bit tracks with what i felt too, just wasn't sure if it was real or in my head. Might look into getting my own levels checked so i'm not just guessing.
  • student748: That's heaps useful actually, 15% total glutathione bump in six weeks is pretty decent for what it is worth. Did you happen to check oxidised vs reduced ratio or just total? Keen to know if the GSH/GSSG balance shifted at all cos that's the part i reckon matters more for recovery than absolute levels. Might get my own panel done next month to compare.
  • student748: Yeah the glycine sleep angle is a, i hadn't really thought about that being a separate variable. For what it is worth i was taking glycine on its own months ago for sleep and did notice something similar, so you might be onto it there. No bloodwork yet, just been going by feel which i know is dodgy. Keen to get glutathione levels checked next time i'm at the gp, see if the glynac actually shifted anything. Did you ever run glynac yourself or just looking at it from the research side?
