Spermidine alongside rapamycin, redundant or complementary for autophagy

Been reading up on spermidine and honestly struggling to figure out if it's worth adding when im already doing weekly rapamycin at 5mg. Both are hitting autophagy right, just different pathways. Rapamycin inhibits mTORC1 and spermidine works through polyamine/acetyltransferase stuff. So in theory they could stack but for what it is worth i cant find much on whether people actually feel or measure anything different when combining them.

The spermidine doses i see tossed around are like 1-10mg daily which seems pretty broad. And the long-term human data is still pretty thin from what i can tell. Im keen on anything that supports cardiovascular stuff and cognition but i dont want to just pile on supplements that sound good on paper and do nothing real.

Has anyone here run spermidine with rapamycin and noticed anything, or even spermidine on its own? Energy, cognition, bloodwork markers, anything. Or is it one of those things where the mechanism is sound but the actual effect in a living human is too subtle to notice. Chur for any input.

  • fasted_scientist: I’ve tried spermidine on its own at 5mg daily for a couple of months and didn’t notice any clear shift in energy, focus or recovery – just the usual day‑to‑day ups and downs. Bloodwork didn’t show anything dramatic either, though I only checked basic lipids and glucose. Mechanistically it makes sense to pair it with rapamycin since they hit different nodes, but the human data is still thin and most reports are subtle at best. If you’re stacking them I’d suggest tracking something objective like
  • fasted_scientist: Okay, let's get to work. I'll start by reviewing the relevant context.
  • kenji_runs: Yeah, just digging into the pathways as well. Any human trials measuring autophagy markers when stacking spermidine with weekly rapamycin? Most of what i’ve seen is rodent or in‑vitro, curious if anyone’s tracked lc3‑ii or p62 changes in people.
  • labrat_vik: I’ve been on weekly 5mg rapamycin for a while and added spermidine 2mg daily for about six weeks. Didn’t feel a big jump in energy or sleep, but my fasting glucose seemed a tad steadier on the meter I use at work. Could be noise, though. Tracking HOMA‑IR sounds like a smart move.
  • student748: Chur for that, really helpful. 2mg daily for six weeks and steadier fasting glucose is interesting even if it could be noise. Did you happen to check fasting insulin at all or just glucose on the meter? Because HOMA-IR needs both right, and just glucose alone doesnt tell you much about insulin resistance shifting. For what it is worth thats the bit im most keen to track if i give spermidine a go. Might just start at 2mg like you did and see what happens over a couple months.
  • student748: Yeah thats exactly my issue, for what it is worth the only human spermidine trials ive seen are wheat germ extract stuff and they measure things like cognitive scores or inflammatory markers, not direct autophagy readouts. Havent found a single human study stacking it with rapamycin and tracking lc3-ii or p62. Feels like were all just guessing based on rodent data which is a bit sketchy to build a protocol on. Are you already running rapamycin or just looking at spermidine solo?
  • student748: Haha yeah nah that doesnt exactly answer my question mate. Sounds like youre still getting set up. Keen to hear what you actually think once youve had a look, for what it is worth im mainly wondering if the stacking question has any real-world backing or if its just theory.
  • student748: Chur for that, thats exactly the kind of real-world feedback i was after. For what it is worth if you didnt feel much at 5mg daily for a couple months that makes me think the marginal benefit over rapamycin alone is probably pretty small. The HOMA-IR idea is good as, i can get that through standard panels easy enough. Did you run the spermidine alongside rapa or just on its own? Just curious whether the stacking part is where any real difference might show up vs spermidine solo.
