Trying PE 22-28 for mood, any logs?

Hey folks, I decided to give PE 22-28 a shot after reading about its TREK-1 blockade and fast antidepressant angle in animal models. I am not a doctor, just sharing my own tbh experience. I started with a very low dose, maybe 100 mcg intranasal once a day, because I have no human data to go on.

First three days I felt nothing special, just a bit of mild headache in the afternoon. On day four I noticed my mood felt a little lighter during my night shift, less irritable after tough calls. Tbh I also felt a bit more mentally clear when I had to write reports, but could be placebo or just better sleep from being less stressed.

I kept a simple log, rating mood 1-10 each evening, and saw a small bump from 5 to 6-7 after day five. I stopped after ten days because I wanted to see if any effect lingered, and after stopping I returned to baseline within two days. If I try again I would maybe split dose twice daily and track sleep more closely.

Anyone else tried this peptide for mood or neurogenesis? Would love to hear your logs or thoughts on dosing.

  • sleep_coastal: I ran PE 22-28 for about two weeks last fall, also intranasal but closer to 200 mcg split morning and evening. Anecdotally the twice-daily split felt smoother than once daily, less of that afternoon headache you mentioned, though mine was more a sinus pressure thing than a true headache. Mood lift for me was subtle too, maybe half a point on a similar scale, and it took about a week to really notice anything. The thing that surprised me was how fast it dropped off after stopping, basically back
  • coastal_labrat: Yeah the dropoff is wild right, like you stop and two days later its gone completely. Kinda makes me wonder if its even doing anything long term or just masking while its active? I did track sleep actually, just my garmin sleep score, and it went from like 65 avg to 72-75 during the run but honestly that could just be the mood lift making me less wound up at bedtime. Did you try cycling it at all or just ran straight through those two weeks?
  • curious_scientist: I tried PE 22-28 intranasal 150 mcg once daily for ten days. I noticed a slight mood lift after day five, but got a mid‑afternoon sinus pressure feeling. Splitting the dose might smooth that out, like you found. Did you see any change in sleep quality?
  • coastal_labrat: I didn’t track sleep closely but felt a bit more rested on nights after my shift, likely from lower stress rather than direct effect. Tbh my headache was mild, not sinus pressure. Splitting the dose helped smooth the headache for me.
