Thoughts on PE 22-28 and the TREK-1 pathway

I was reading up on the mechanism for PE 22-28 (/peptides/pe-22-28) and the way it blocks the TREK-1 potassium channel. The idea that it could trigger synaptogenesis and neurogenesis faster than standard SSRIs is class.

Fwiw, I have always been more interested in the structural side of brain health than just masking symptoms. The preclinical data on rapid antidepressant effects seems promising, but I wonder if we are overhyping the speed of it for humans.

Most of the stuff I have tried for cognitive health takes weeks to really settle in (if it works at all). If this actually delivers a fast shift in mood or plasticity, it would be a game changer. That said, it is still preclinical, so I am a bit skeptical about how those animal models translate to us.

It feels a bit too good to be true, but I would love to see some actual human logs on it. Do you think the TREK-1 approach is actually a better route for longevity and brain health than the usual nootropics? 🧠
