PE 22-28 and the TREK-1 channel research

I was reading some recent papers on TREK-1 potassium channel blockers and it basically brings me back to the interest in PE 22-28. This peptide is a derivative of spadin and it is designed to block those specific channels to boost serotonergic signaling. The preclinical data suggesting it might work much faster than traditional SSRIs for neurogenesis is very interesting.
For me, the speed of action is the most fascinating part. Most antidepressant research focuses on slow changes in receptor density, but the idea of rapid synaptogenesis through TREK-1 inhibition seems like a different mechanism entirely. However, since it is still in the preclinical stage, I am very skeptical about how it will actually translate to human subjects. We do not have any established human safety data or reliable dosing protocols yet.
I am curious if anyone else is following the specific progress on spadin derivatives. Do you think the potential for rapid neuroplasticity outweighs the massive unknown risks regarding how we might alter background potassium channels in the brain? It seems like a very high risk, high reward area of research.
