Trying a Combined Rapamycin/Spermidine Protocol – How’s Yours Going?

I’ve been on low‑dose rapamycin (4mg once a week) for about three months now, switching to a 2‑week cycle to keep my immune system from getting too suppressed. The energy boost was real – I could still hit the gym after a full shift on the fire brigade, but my sleep quality did dip a bit and I’ve noticed mild mouth sores that pop up after a couple of weeks on the drug. I’ve been adding 5mg of spermidine daily to try to kick‑start autophagy and maybe offset the GI upset. The spermidine has been easy to take, a small capsule with water, and I’ve not felt any new side effects.
I’d love to hear if anyone else is mixing these two or has tried a different rapamycin schedule. How do you feel the cycle length affects your sleep and energy? Also, any tips for managing the mouth sores without messing up the rapamycin levels?

- liam_sleepnerd: I’m on a similar track – 4 mg weekly rapamycin with a 2‑week break, and 3 mg spermidine daily. The 2‑week off cycle helps keep my HRV up and sleep a touch more regular than when I went straight 7ocular. The mouth sores show up after about 3–4 weeks on rap, so I started a simple salt‑water rinse after each meal and use a soft toothbrush. It keeps the lesions small and doesn’t seem to alter serum rap levels when I’ve checked. Just a heads‑up that the rinse can be a bit gritty for some people, so
- greg208: Got it, not gonna lie rinse sounds handy. I’ve seen sores after the third week, so I’ll try that too. How’s your HRV on the 2‑week off days versus the on days? Any tweaks you do to keep that post‑shift lift steady?
- hank_m: I’ve also been doing a salt‑water rinse after meals and it really helps keep the lesions from getting big, tbh the gritty feel is the only hiccup, but the rinse doesn’t mess with my rap levels like you said. I still get that post‑shift lift but my HRV’s a touch steadier on the off weeks. What’s the humidity like in your kitchen?
- greg208: Thanks for the tip on the salt‑water rinse, I’ve tried it but the gritty taste was a bit brutal. The humidity in my kitchen is around 45 % RH, with a small humidifier running the night – the fridge stays cooler though. HRV still feels a bit wobbly on the rap weeks, maybe my 2‑week cycle is a bit long. I’ll give the rinse a go and see if the lesions stay smaller without messing with the drug. Any other tricks to ease the grit?
