First Time Trying Weekly Rapamycin – Thoughts & Side Effects?

Posted by grace_sleepnerd in Longevity & Anti-Aging - 1 points, 3 comments.

I'm new to the whole longevity stack thing, so I’ve just started the 3 mg once‑weekly rapamycin that the telemed folks prescribed. Been reading about the autophagy angle and how it mimics caloric restriction. I got my first dose last Tuesday, and the next day I felt a bit sore in my mouth, which I’m guessing is a common side effect.

I don't have any major health issues, but I’m watching the lipids in my blood. I’m also on metformin 500 mg twice a day for blood sugar control, so I was wondering if anyone has tried the combo and what they saw in their cholesterol or immune markers. I’m hoping for maybe a boost in energy over the next few weeks, but I know it can take time to see changes.

If y’all have any tips on monitoring or if you felt any other side effects, let me know. Thanks!

  • chemist_luis: I’ve been on weekly rapamycin for about 3 months now. The first dose usually gives that mouth‑sore you mentioned, like a dry, slightly irritated surface. I tied it to a light mucus change, nothing serious. I’ve been checking my lipids every 4 weeks; i hit a small drop in LDL and a slight rise in HDL, about 5‑ trợ. No big shifts, but it’s a trend worth tracking. Combining it with metformin? I’ve had that combo too. The blood sugar stayed steady, and the triglycerides went down a bit; no immune m
  • hank_m: Sounds like your numbers are in the same ballpark as mine – a modest LDL dip and HDL bump after a few weeks, and that IL‑6 spike that pops up just after the dose. I’ve had metformin on the side too, and glucose stayed flat, but i did notice a small drop in triglycerides after a month of weekly rapamycin, probably the same effect you saw. Just keep an eye on the lipids and maybe grab a CRP in 3 months to see if the inflammation settles. Any other side‑effects you’re watching?
  • hank_m: Tbh chemist_luis i’ve been on the same 3 mg weekly rapamycin for six weeks while staying on metformin 500 mg bd and i noticed the mouth‑sore only the first two doses then it settled, my LDL dropped about 8 points and triglycerides slipped a tad, HDL stayed flat, no big immune shifts i could feel but i’m keen to keep tracking the lipids like you do tbh.
