I came across a pre‑clinical paper on PE‑22‑28 last week – the same spadin‑derived peptide that blocks the TREK‑1 channel and, in mice, is shown to lift mood in a few hours insteaduves of weeks for SSRIs. The authors link the effect to increased serotonergic signalling and a burst of synaptogenesis. It’s striking, but the paper emphasises that it’s still research‑grade and we have no idea about human safety or dosing.
For me, the idea that a single dose could give a rapid lift is tempting, but I remain skeptical until there are human trials or even a small safety study. In the meantime, I’ve been sticking to more established peptide stacks, like BPC‑157 for rehab and a low‑dose Metformin for energy. If anyone has tried any peptides that target ion channels or serotonin in a similar way, or if someone’s following a protocol that might hint at mood changes, I’d love to hear your thoughts. Could a low‑dose tweak of PE‑22‑28 be a realistic short‑term mood aid, or is it just hype at this point?
